謝建台 教授 (Prof. Shiea Jentaie)

簡介

謝建台教授於1980年畢業於中興大學化學系,19881991年分別取得美國蒙大拿州立大學 (Montana State University, MSU) 有機地球化學碩士及分析化學博士學位,隨後赴賓州州立大學 (Pennsylvania State University, PSU) 材料科學系擔任博士後研究,再於1991年回到國立中山大學化學系任教迄今。

研究方向主要有三:
(1)新大氣壓力質譜游離源之開發,以及其在食品安全、鑑識科學、環境化學、工業化學、醫藥學、反恐、及動植物化學等相關領域之應用
(2)高效液相層析、氣相層析、毛細管電泳、以及薄層層析-質譜介面的開發
(3)以現代質譜法快速偵測生物體液中之各式疾病生物指標以及應用質譜分子影像技術於生物醫學科學

過去十年來,他積極參與並致力於推動台灣以及亞洲區的質譜研究,曾擔任世界質譜基金會 (International Mass Spectrometry Foundation, IMSF) 執行理事兼亞太區代表及台灣質譜學會會長。他從事質譜研究多年,曾獲得多項國內外學術研究獎項如:2009年行政院國家科學委員會所頒發之『傑出研究獎』、2010年行政院經濟部頒發之『國家創作發明獎』、2011年中國化學會頒發的『化學技術獎章』、2012年由遠東集團有庠基金會所頒發之『有庠科技發明獎』、2013年由美國質譜學會所頒發之『傑出論文審查人獎』、由台灣質譜學會所頒發之『優秀質譜研究學者獎』以及 2017 年由行政院科技部所頒發之『傑出技術移轉貢獻獎』。在國立中山大學服務迄今,分別於 20022006 2009年獲得『國立中山大學研究傑出獎』,以及 200720112017年的『中山發明獎』及『產學傑出獎』等獎項。2010年也獲英國皇家化學會頒發英國皇家化學會會士頭銜。他也曾擔任多項政府單位之審查、審議及諮議委員,也是國立中山大學學術研究及產學研究兩項之特聘教授,除此之外現在也是 J. Am. Soc. Mass Spectrom. (JASMS),  Clin. Mass Spectrom. (CMS), Mass Spectrom., Mass Spectrom. Let., Int. J. Mass Spectrom. (IJMS), Rapid Commun. Mass Spectrom. (RCM), Anal. Methods, Technology, Clin. 此技術是將具熱不穩定性及高揮發性之化合物溶於低溫的有機溶劑來抑制其化學活性,並通過低溫管路導入到 ESI 游離源或是二次離子源中進行游離。相較於傳統分析法,此技術可得更佳的不穩定分析物離子訊號。

此技術是施加高電壓於一上面置有一微滴之樣品溶液之金屬絲上,金屬絲表面可以是經修飾(如奈米氧化鎢管) 或不經修飾 (如銅絲)。樣品液滴在受到高電場的影響下,會移向質量分析器並在金屬絲尖端產生微電噴灑現象。此技術因組裝簡單,使用材料便宜,樣品更換容易,因此未來可應用於大量樣品數目的分析。

瞬間熱裂解 (Pyrolysis, Py) 結合融合微滴電噴灑游離法 (Fused-droplet electrospray ionization, FD-ESI)   電噴灑輔助雷射脫附游離法 (Electrospray-Assisted Laser Desorption Ionization, ELDI)

FD-ESI 是我們實驗室所開發出來的大氣壓力游離技術。我們發現在將 Py 結合 FD-ESI 後,我們可以快速偵測到來自高分子材料的熱裂解極性產物,以此方法,我們可以很容易區分各式不同高分子材料。我們發現儘管蛋白質溶液中含有大量的有機或是無機鹽類,但均可在不經任何樣品前處理步驟下,以FD-ESI/MS 直接得到蛋白質離子的訊號。和傳統的ESI/MS 分析結果比較起來,此技術對鹽類的容忍度是前者的100倍以上。

薄層層析法 (Thin Layer Chromatography, TLC) 具有操作簡單、成本低廉、樣品需求量少的特性,且已被廣泛應用在小分子的分析上,與電噴灑的結合可提供直接且快速地沖提物檢測。而電噴灑輔助雷射脫附游離法 (Electrospray Laser Desorption Ionization, ELDI) 可在一大氣壓力下,不用複雜的樣品前處理,就直接對樣品進行快速的分析。本研究是利用市售樂高模組具可輕易組裝的優點,並與輸送帶結合,發展成一套高通量且可快速篩選各式成分的TLC-ELDI/MS偵測系統,可針對大量樣品包含藥物、化妝品等進行高效率的分析。

我們將蛋白質溶液塗敷於一鋁金屬薄片上,鋁膜另一面再以雷射照射,以產生震波造成蛋白質分子的脫附,這些被脫附之蛋白質分子在移動到一電噴灑區後,會進一步被電噴灑所游離。我們發現不論樣品是以固體或是液體,均可以此法成功被游離。

熱脫附電噴灑游離質譜法 (Thermal Desorption Electrospray Ionization, TD-ESI)   雙電噴灑及大氣壓化學游離法 (Dual Electrospray and Atmospheric Pressure Chemical Ionization)

熱脫附電噴灑游離源技術包含了熱脫附裝置和電噴灑離子源,利用一金屬取樣探針沾取或刮取樣品,可於開放空間下分析固體、液體、凝膠及膏狀等型態的樣品,經由加熱區以高溫加熱脫附樣品,並以預先加熱的氮氣作為載流氣體,將分析物攜帶至電噴灑噴霧中產生游離反應,全程只需約 30 秒便可知道結果。具有方便、簡單、快速、即時且感度高等優點。此分析技術提供不同領域的分析者檢測多種不同基質的樣品或於其表面做取樣分析;如尿液中安非他命系列的檢測。

開發了結合電噴灑游離(ESI)和大氣壓化學游離(APCI)的雙游離源,同時電離極性和非極性化合物。將高直流電壓施加到在熔融石英毛細管中流動的甲醇溶液中,在以毛細管尖端處產生ESI電噴灑。將高交流電壓施加到連接到玻璃管的環形電極,在玻璃管與不銹鋼管柱間流動的氮氣產生電漿。由於ESIAPCI所需的高電壓是獨立應用和控制的,因此雙離子源可以在僅ESI,僅APCIESI + APCI模式下運行。通過用連續波長激光或脈衝激光束照射樣品表面將分析物引入ESIAPCI流中。通過將雙ESI+APCI向樣品表面以進行脫附和游離,也可以產生分析物離子。雙離子源中涉及的游離機制包括潘寧電離,離子分子反應和熔滴電噴霧電離。

火焰誘導大氣壓化學游離法 (Flame-Induced Atmospheric Pressure Chemical Ionization, FAPCI)   多通道電噴灑游離法 (Multiple Channel Electrospray Ionization)

火焰誘導的大氣壓化學游離(FAPCI)是一種溶劑和高電壓組合的 APCI 技術。它利用火焰產生帶電物質,與分析物反應進行游離,並從有機化合物中生成完整的分子離子,與較少的碎片離子。在這項研究中,脫附 FAPCI/MS 被開發用於快速偵測液態,糊狀物和固態的熱穩定有機化合物。通過加熱的噴灑器將液體樣品引入離子源,並且在加熱的噴霧器中形成的分析物與游離源中的帶電物質反應。對於糊狀物和固體樣品分析,會將樣品放置於游離源的火焰附近以進行熱脫附和游離。

旋轉電噴灑游離質譜法 (Rotating Electrospray Ionization, RESI) 可用來有效提升離子-分子反應 (Ion-Molecule Reaction, IMR) 的效率。其是藉由機動式馬達將不斷噴出帶電液滴的電噴灑游離源進行高速旋轉,並形成一均勻分布的電噴灑噴霧區域,而此現象特別有益於二段式大氣壓力游離質譜法如 FD-ESI, ELDILIAD-ESI 等技術。除此之外,此技術更可運用在多管式電噴灑游離源上,藉以產生一均勻分布的多管式電噴灑噴霧區域,以克服靜止式因電場干擾所造成的電噴灑不穩定及電噴灑噴霧不均勻等問題。

生物醫學 (Biomedicine):

乳癌指標 (Breast Cancer)

乳癌是因為乳房乳腺管細胞或是乳小葉細胞產生不正常分裂、繁殖而形成之惡性腫瘤。乳癌除了局部侵犯乳房組織外,更可能轉移到遠處組織,如:肺、骨骼、肝臟、腦等器官,而危害生命安全。
  糞便潛血指標 (Fecal Occult Blood, FOB)

大腸癌的腫瘤或息肉會受到糞便形成過程中的摩擦而流血,血液因而混合在糞便內排出,使得糞便表面含有肉眼看不出來的血液,透過糞便潛血檢查可以早期發現大腸癌或息肉等症狀。
  蛋白尿指標 (Albuminuria)

一般正常尿液因排泄系統構造的關係,尿中蛋白質應小於150毫克 (白蛋白每日排出量應小於30毫克),尿中蛋白質高於標準值時,可能導因於是否有腎臟或心臟方面的疾病。

是一種以表現在行為、心理活動上的紊亂為主的精神症狀。其原因可能來自外在因素如家庭、社會環境,或是患者自身的生理遺傳因素、神經生化因素等內在原因相互作用所導致的心理活動、行為、及其神經系統功能紊亂,目前臨床診斷較難有具體的疾病指標判別。
  防禦素發炎指標 (Defensins)

人體免疫系統的細胞含有這些防禦素協助殺死被吞噬的細菌,具有很強的抗細菌、真菌和具外套膜病毒作用,其存於嗜中性粒細胞和幾乎所有的上皮細胞。

胃幽門桿菌毒性鑑定 (Bacteria Identification) 

此菌的螺旋結構及菌上頭的鞭毛,使它能夠鑽入胃黏液而達到胃黏膜上,在黏液與黏膜之間酸度是中性的,而且此細菌本身也可以分泌尿素脢來保護自己,在胃裡面會破壞胃黏膜造成胃發炎,從急性的胃炎到慢性的胃炎它都可以一肩挑,擔任相當吃重的角色,目前已確定胃炎的重要罪魁禍首之一就是它。
  糖化血色素指標 (HbA1c)

糖化血色素是最容易反映血糖濃度的指標之一,血液中的葡萄糖濃度愈高,累積的HbA1c濃度與比率也愈高,且因為紅血球的平均壽命約為3個月,所以測定血液HbA1c的百分比,就可以估計過去120天期間內的平均血糖濃度,目前是糖尿病患者控制血糖的最佳指標。一般來說,HbA1c的正常檢測參考值約為4~6%,國際間採用之糖尿病治療準則是將HbA1c控制在7%以下。
  影像質譜法 (Mass Spectrometry Imaging , MSI)

此分析方法可利用質譜訊號來分析物質組成,更可了解特定物質在空間中的分布情形。此分析方法之優勢即是不需要使用任何標記或抗體標誌物,即可直接在組織樣品上進行分子影像觀察與研究,並了解其分子空間分布資訊,且能找出與腫瘤發生相關之生化分子。
